That’s a real worry, especially because these chemicals might change into something even worse after water disinfection. So, we decided to look into the biological activity of eight common EDCs and PPCPs found in source water, both before and after disinfection. These included acetaminophen, bisphenol A, carbamazepine, estrone, 17 alpha-ahinylestradiol, gemfibrozil, naproxen, and triclosan.<\/p>\n<p class=\"FR_fieldWe used a mix of computer modeling and lab experiments for this study. With a method called Stepped Forced Molecular Dynamics (SFMD), we found 911 unique transformation products, and it turns out we've seen 36% of those in other scientific reports. We also figured out how likely these transformation products were to harm biomolecules or DNA compared to the original compounds. We based this on how well they dissolve in fat and if they had certain \"danger\" structures. Plus, we used two tools to predict their toxicity through how they interact with cell receptors.<\/p>\n<p class=\"FR_fieldIn the lab, we ran experiments with three different disinfectants: chlorine, chlorine dioxide, and chloramine. After we pulled out the solids, we checked the resulting mixtures of transformation products using chemical analysis and eleven different in vitro bioassays. These bioassays looked at a bunch of different effects. What we saw in the lab matched our predictions pretty well. All in all, our combined approach of computational and experimental chemistry and toxicity suggests that disinfecting these EDCs and PPCPs will create a lot of new transformation products. While they might not boost specific toxicities, like endocrine activity, they could increase general reactive and non-specific toxicity. (C) 2018 Elsevier B.V.
